Amyloidosis Glossary of Terms

The following are health and medical definitions of terms that appear in the Amyloidosis article.

Abdominal: Relating to the abdomen, the belly, that part of the body that contains all of the structures between the chest and the pelvis. The abdomen is separated anatomically from the chest by the diaphragm, the powerful muscle spanning the body cavity below the lungs.

Abnormal: Not normal. Deviating from the usual structure, position, condition, or behavior. In referring to a growth, abnormal may mean that it is cancerous or premalignant (likely to become cancer).

Aging: The process of becoming older, a process that is genetically determined and environmentally modulated.

Alzheimer's disease: A progressive neurologic disease of the brain that leads to the irreversible loss of neurons and dementia. The clinical hallmarks of Alzheimer's disease are progressive impairment in memory, judgment, decision making, orientation to physical surroundings, and language. A working diagnosis of Alzheimer disease is usually made on the basis of the neurologic examination. A definitive diagnosis can be made only at autopsy. On a cellular level, Alzheimer's disease is characterized by unusual helical protein filaments in nerve cells (neurons) of the brain. These odd twisted filaments are called neurofibrillary tangles. On a functional level, there is degeneration of the cortical regions, especially the frontal and temporal lobes, of the brain.

Amyloid: Any of a number of complex proteins that are deposited in tissues and that share selected laboratory features such as a change in the fluorescence intensity of certain aromatic dyes like Congo Red.

Amyloidosis: A disorder that results from the abnormal deposition of a particular protein, called amyloid, in various tissues of the body. Amyloid protein can be deposited in a localized area, and not be harmful, or in can cause serious changes in virtually any organ of the body.

Ankylosing: Undergoing ankylosis (stiffening or fusion of a joint).

Ankylosing spondylitis: A type of arthritis that causes chronic inflammation of the spine and the sacroiliac joints. Chronic inflammation in these areas causes pain and stiffness in and around the spine. Over time, chronic spinal inflammation (spondylitis) can lead to a complete cementing together (fusion) of the vertebrae, a process called ankylosis. Ankylosis causes total loss of mobility of the spine.

Antibody: An immunoglobulin, a specialized immune protein, produced because of the introduction of an antigen into the body, and which possesses the remarkable ability to combine with the very antigen that triggered its production.

Arthritis: Inflammation of a joint. When joints are inflamed they can develop stiffness, warmth, swelling, redness and pain. There are over 100 types of arthritis. (see osteoarthritis, rheumatoid arthritis, ankylosing spondylitis, psoriatic arthritis, lupus, gout, pseudogout).

Aspiration: Removal of a sample of fluid and cells through a needle. Aspiration also refers to the accidental sucking in of food particles or fluids into the lungs.

Autosomal: Pertaining to a chromosome that is not a sex chromosome; relating to any one of the chromosomes save the sex chromosomes.

Autosomal dominant: A pattern of inheritance in which an affected individual has one copy of a mutant gene and one normal gene on a pair of autosomal chromosomes. (In contrast, autosomal recessive diseases require that the individual have two copies of the mutant gene.) Individuals with autosomal dominant diseases have a 50-50 chance of passing the mutant gene and therefore the disorder onto each of their children. Examples of autosomal dominant diseases include Huntington disease, neurofibromatosis, and polycystic kidney disease.

Belly: That part of the body that contains all of the structures between the chest and the pelvis. Also called the abdomen.

Biopsy: The removal of a sample of tissue for purposes of diagnosis. (Many definitions of "biopsy" stipulate that the sample of tissue is removed for examination under a microscope. This may or may not be the case. The diagnosis may be achieved by other means such as by analysis of chromosomes or genes.)

Bone marrow: The soft blood-forming tissue that fills the cavities of bones and contains fat and immature and mature blood cells, including white blood cells, red blood cells, and platelets. Diseases or drugs that affect the bone marrow can affect the total counts of these cells.

Brain: That part of the central nervous system that is located within the cranium (skull). The brain functions as the primary receiver, organizer and distributor of information for the body. It has two (right and left) halves called "hemispheres."

Cancer: An abnormal growth of cells which tend to proliferate in an uncontrolled way and, in some cases, to metastasize (spread).

Cell: The basic structural and functional unit in people and all living things. Each cell is a small container of chemicals and water wrapped in a membrane.

Chemotherapy: 1. In the original sense, a chemical that binds to and specifically kills microbes or tumor cells. The term chemotherapy was coined in this regard by Paul Ehrlich (1854-1915).
2. In oncology, drug therapy for cancer. Also called "chemo" for short.

Clinical trials: Trials to evaluate the effectiveness and safety of medications or medical devices by monitoring their effects on large groups of people.

Colchicine: A substance found in a plant that is used in clinical medicine for the treatment of gouty arthritis and in the laboratory to arrest cells during cell division (by disrupting the spindle) so their chromosomes can be visualized. The name colchicine is from the Greek kolchikon meaning autumn crocus or meadow saffron, the plant from which colchicine was originally isolated.

Diagnosis: 1 The nature of a disease; the identification of an illness. 2 A conclusion or decision reached by diagnosis. The diagnosis is rabies. 3 The identification of any problem. The diagnosis was a plugged IV.

Dialysis: The process of cleansing the blood by passing it through a special machine. Dialysis is necessary when the kidneys are not able to filter the blood. Dialysis allows patients with kidney failure a chance to live productive lives. There are two types of dialysis: hemodialysis and peritoneal dialysis. Each type of dialysis has advantages and disadvantages. Patients can often choose the type of long term dialysis that best matches their needs.

Familial: A condition that is tends to occur more often in family members than expected by chance alone. A familial disease may be genetic (such as cystic fibrosis) or environmental (such as tuberculosis).

Fatigue: A condition characterized by a lessened capacity for work and reduced efficiency of accomplishment, usually accompanied by a feeling of weariness and tiredness. Fatigue can be acute and come on suddenly or chronic and persist.

Fever: Although a fever technically is any body temperature above the normal of 98.6 degrees F. (37 degrees C.), in practice a person is usually not considered to have a significant fever until the temperature is above 100.4 degrees F (38 degrees C.).

Genetics: The scientific study of heredity. Genetics pertains to humans and all other organisms. So, for example, there is human genetics, mouse genetics, fruitfly genetics, etc.

Heart: The muscle that pumps blood received from veins into arteries throughout the body. It is positioned in the chest behind the sternum (breastbone; in front of the trachea, esophagus, and aorta; and above the diaphragm muscle that separates the chest and abdominal cavities. The normal heart is about the size of a closed fist, and weighs about 10.5 ounces. It is cone-shaped, with the point of the cone pointing down to the left. Two-thirds of the heart lies in the left side of the chest with the balance in the right chest.

Incidence: The frequency with which something, such as a disease, appears in a particular population or area. In disease epidemiology, the incidence is the number of newly diagnosed cases during a specific time period. The incidence is distinct from the prevalence which refers to the number of cases alive on a certain date.

Infection: The growth of a parasitic organism within the body. (A parasitic organism is one that lives on or in another organism and draws its nourishment therefrom.) A person with an infection has another organism (a "germ") growing within him, drawing its nourishment from the person.

Inflammation: A basic way in which the body reacts to infection, irritation or other injury, the key feature being redness, warmth, swelling and pain. Inflammation is now recognized as a type of nonspecific immune response.

Injury: Harm or hurt. The term "injury" may be applied in medicine to damage inflicted upon oneself as in a hamstring injury or by an external agent on as in a cold injury. The injury may be accidental or deliberate, as with a needlestick injury. The term "injury" may be synonymous (depending on the context) with a wound or with trauma.

Joint: A joint is the area where two bones are attached for the purpose of motion of body parts. A joint is usually formed of fibrous connective tissue and cartilage. An articulation or an arthrosis is the same as a joint.

Kidney: One of a pair of organs located in the right and left side of the abdomen which clear "poisons" from the blood, regulate acid concentration and maintain water balance in the body by excreting urine. The kidneys are part of the urinary tract. The urine then passes through connecting tubes called "ureters" into the bladder. The bladder stores the urine until it is released during urination.

Liver: An organ in the upper abdomen that aids in digestion and removes waste products and worn-out cells from the blood. The liver is the largest solid organ in the body. The liver weighs about three and a half pounds (1.6 kilograms). It measures about 8 inches (20 cm) horizontally (across) and 6.5 inches (17 cm) vertically (down) and is 4.5 inches (12 cm) thick.

Lungs: The lungs are a pair of breathing organs located with the chest which remove carbon dioxide from and bring oxygen to the blood. There is a right and left lung.

Marrow: The bone marrow.

Mediterranean Fever: A inherited disorder of unknown cause featuring short recurring bouts of fever together with pain in the joints, chest or abdomen. Also called Familial Mediterranean Fever (FMF).

Mouth: 1. The upper opening of the digestive tract, beginning with the lips and containing the teeth, gums, and tongue. Foodstuffs are broken down mechanically in the mouth by chewing and saliva is added as a lubricant. Saliva contains amylase, an enzyme that digests starch. 2. Any opening or aperture in the body. The mouth in both senses of the word is also called the os, the Latin word for an opening, or mouth. The o in os is pronounced as in hope. The genitive form of os is oris from which comes the word oral.

Multiple myeloma: A malignancy of plasma cells (a form of lymphocyte) that typically involves multiple sites within the bone morrow and secretes all or part of a monoclonal antibody . Also called plasma cell myeloma.

Myeloma: A tumor of antibody-producing cells, called plasma cells, that are normally found in the bone marrow.

Organ: A relatively independent part of the body that carries out one or more special functions. The organs of the human body include the eye, ear, heart, lungs, and liver.

Organ failure: The failure of an essential system in the body. Multiple organ failure is the failure of two or more systems, such as the cardiovascular, and renal systems, and is a common consequence of sepsis (the presence of bacteria in the bloods) and of shock (very low blood pressure).

Osteomyelitis: Inflammation of the bone due to infection, for example by the bacteria salmonella or staphylococcus. Osteomyelitis is sometimes a complication of surgery or injury, although infection can also reach bone tissue through the bloodstream. Both the bone and the bone marrow may be infected. Symptoms include deep pain and muscle spasms in the area of inflammation, and fever. Treatment is by bed rest, antibiotics (usually injected locally), and sometimes surgery to remove dead bone tissue.

Plasma: The liquid part of the blood and lymphatic fluid, which makes up about half of its volume. Plasma is devoid of cells and, unlike serum, has not clotted. Blood plasma contains antibodies and other proteins. It is taken from donors and made into medications for a variety of blood-related conditions. Some blood plasma is also used in non-medical products.

Plasma cell: A type of white blood cell that produces and secretes antibodies. A plasma cell is a fully differentiated lymphocyte in the B-cell (not the T-cell) lineage. As with most cell types, plasma cells can mutate to give rise to cancer. Plasma cell malignancies include multiple myeloma, Waldenstrom macroglobulinemia, and plasma cell leukemia.

Primary amyloidosis: A disorder of plasma cells (special white blood cells that produce antibodies), this is one of a group of diseases (called amyloidosis) in which protein deposits (amyloid) accumulate in one or more organ systems in the body. Primary amyloidosis is the most common type of amyloidosis in many countries including the U.S.

Rectum: The last 6 to 8 inches of the large intestine. The rectum stores solid waste until it leaves the body through the anus. The word rectum comes from the Latin rectus meaning straight (which the human rectum is not).

Rheumatoid arthritis: An autoimmune disease which causes chronic inflammation of the joints, the tissue around the joints, as well as other organs in the body. Autoimmune diseases occur when the body tissues are mistakenly attacked by its own immune system. The immune system is a complex organization of cells and antibodies designed normally to "seek and destroy" invaders of the body, particularly infections. Patients with these diseases have antibodies in their blood which target their own body tissues, where they can be associated with inflammation. Because it can affect multiple other organs of the body, rheumatoid arthritis is referred to as a systemic illness and is sometimes called rheumatoid disease. While rheumatoid arthritis is a chronic illness (meaning it can last for years) patients may experience long periods without symptoms.

Secondary amyloidosis: One of a group of diseases (called amyloidosis) in which protein deposits (amyloid) accumulate in one or more organ systems in the body, secondary amyloid is caused by a chronic infection or inflammatory disease such as rheumatoid arthritis, familial Mediterranean fever, osteomyelitis, or granulomatous ileitis.

Shortness of breath: Difficulty in breathing. Medically referred to as dyspnea. Shortness of breath can be caused by respiratory (breathing passages and lungs) or circulatory (heart and blood vessels) conditions. See also dyspnea.

Spondylitis: Inflammation of one or more of the vertebrae of the spine. Diffuse inflammation of the spine is seen in the disease ankylosing spondylitis. Localized spondylitis is seen with infections of a certain area of the spine, such as in Pott's disease.

Stem cells: One of the human body's master cells, with the ability to grow into any one of the body's more than 200 cell types.

Syndrome: A set of signs and symptoms that tend to occur together and which reflect the presence of a particular disease or an increased chance of developing a particular disease.

Systemic: Affecting the entire body. A systemic disease such as diabetes can affect the whole body. Systemic chemotherapy employs drugs that travel through the bloodstream and reach and affect cells all over the body.

Tongue: The tongue is a strong muscle anchored to the floor of the mouth. It is covered by the lingual membrane which has special areas to detect tastes.

Transthyretin: A plasma protein consisting of 127 amino acids that binds retinol and thyroxine. The gene TTR that encodes transthyretin is in chromosome region 18q11.2-q12.1. Many distinct forms of amyloidosis have been related to different point mutations in TTR. Transthyretin is also called prealbumin (because it migrates faster than albumin in an acidic starch gel).

Tuberculosis: A highly contagious infection caused by the bacterium called Mycobacterium tuberculosis. Abbreviated TB. Tubercles (tiny lumps) are a characteristic finding in TB. Diagnosis may be made by skin test, which if positive should will be followed by a chest X-ray to determine the status (active or dormant) of the infection. Tuberculosis is more common in people with immune system problems, such as AIDS, than in the general population. Treatment of active tuberculosis is mandatory by law in the US, and should be available at no cost to the patient through the public health system. It involves a course of antibiotics and vitamins that lasts about six months. It is important to finish the entire treatment, both to prevent reoccurrence and to prevent the development of antibiotic-resistant tuberculosis. Most patients with tuberculosis do not need to be quarantined, but it is sometimes necessary.

Urine: Liquid waste. The urine is a clear, transparent fluid. It normally has an amber color. The average amount of urine excreted in 24 hours is from 40 to 60 ounces (about 1,200 cubic centimeters). Chemically, the urine is mainly an aqueous (watery) solution of salt (sodium chloride) and substances called urea and uric acid. Normally, it contains about 960 parts of water to 40 parts of solid matter. Abnormally, it may contain sugar (in diabetes), albumen (a protein) (as in some forms of kidney disease), bile pigments (as in jaundice), or abnormal quantities of one or another of its normal components.

Weight loss: Weight loss is a decrease in body weight resulting from either voluntary (diet, exercise) or involuntary (illness) circumstances. Most instances of weight loss arise due to the loss of body fat, but in cases of extreme or severe weight loss, protein and other substances in the body can also be depleted. Examples of involuntary weight loss include the weight loss associated with cancer, malabsorption (such as from chronic diarrheal illnesses ), and chronic inflammation (such as with rheumatoid arthritis).

      • Osteomyelitis is an infection of the bone. Potential causes include injections around the bone, fractures that puncture the skin, recent surgeries, and bacterial infections that travel from other areas of the body, spreading through the blood to the bone. Symptoms include pain, fever, chills, stiffness, and nausea. Treatment involves antibiotics and pain medications. Surgery is sometimes necessary.

      • Orthostatic hypotension symptoms include lightheadedness, weakness, blurred vision, and syncope or passing out. Causes of orthostatic hypotension include dehydration, anemia, medication, blood loss, low blood pressure, heat related illnesses and more. Treatment of orthostatic hypotension depends on the underlying cause.
    • Sudden Cardiac Arrest
      • Sudden cardiac arrest is an unexpected, sudden death caused by sudden cardiac arrest (loss of heart function). Symptoms of brain lesions include headache, nausea, fever, neck pain and stiffness, affected vision and speech, weakness or paralysis to one side of the body. Diagnosis of brain lesions is generally with imaging studies like CT or MRI scans. Treatment and prognosis of brain lesions depends on the cause of the lesion.

Amyloidosis

What is osteomyelitis?

Osteomyelitis is infection in the bone. Osteomyelitis can occur in infants, children, and adults. Different types of bacteria typically affect the different age groups. In children, osteomyelitis most commonly occurs at the ends of the long bones of the arms and legs, affecting the hips, knees, shoulders, and wrists. In adults, it is more common in the bones of the spine (vertebrae) or in the pelvis.

What causes osteomyelitis?

There are several different ways to develop osteomyelitis. The first is for bacteria to travel through the bloodstream (bacteremia) and spread to the bone, causing an infection. This most often occurs when the patient has an infection elsewhere in the body, such as pneumonia or a urinary tract infection, that spreads through the blood to the bone.
